      目的:建立茶芎、川芎、藁本的超高效液相色谱(UPLC)多成分同步定量分析方法,系统剖析三者化学成分差异,明确品种特征标志物,阐释成分差异与药材基原、功效的内在联系。方法:采用 Waters CORTECS T3 C18 色谱柱(150 mm×2.1 mm,1.6 μm),以乙腈 - 0.1% 磷酸溶液为流动相进行梯度洗脱,流速 0.4 mL/min,柱温 30 ℃,检测波长为 230 nm、280 nm,进样量 5 μL;采用外标法同步测定 42 批茶芎、18 批川芎、20 批藁本共 80 批样品中 10 种主要活性成分的含量。结果: 所建立的 UPLC 方法准确可靠,专属性、重复性良好,可实现 10 种成分同步定量。三类药材在生物碱、酚酸、苯酞类物质的整体分布、单体成分含量上存在显著区别,川芎嗪、洋川芎内酯 A、丁苯酞分别为川芎、茶、藁本的特征性成分。结论:茶芎、川芎、藁本化学成分差异显著,不同类别活性成分的分布特征是区分三者的核心依据,也为三者功效分化提供物质基础。

      Objective To establish an ultra-high performance liquid chromatography (UPLC) method for simultaneous quantitative analysis of multi-components in Ligusticum sinense Oliv.cv.Chaxiong, Ligusticum chuanxiong Hort. and Ligusticum sinense Oliv., systematically analyze the differences of their chemical components, identify variety-specific markers, and explore the relationship between component characteristics and medicinal properties. Methods The separation was performed on a Waters CORTECS T3 C18 column (150 mm×2.1 mm, 1.6 μm) with gradient elution using acetonitrile-0.1% phosphoric acid solution as the mobile phase. The flow rate was 0.4 mL/min, the column temperature was 30 ℃, the detection wavelengths were 230 nm and 280 nm, and the injection volume was 5 μL. The contents of 10 main active components in 80 batches of samples (42 batches of L. sinense cv.Chaxiong, 18 batches of L. chuanxiong, 20 batches of L. sinense) were simultaneously determined by external standard method. Results The established UPLC method was accurate, reliable and reproducible. Obvious differences were observed in the distribution of alkaloids, phenolic acids and phthalides among the three medicinal materials. Ligustrazine, senkyunolide A and n-butylphthalide were respectively the characteristic components of L. chuanxiong, L. sinense cv.Chaxiong, and L. sinense. Conclusion There are significant differences in chemical compositions among the three herbs. The distribution characteristics of different active components can be used to distinguish varieties, and lay a material foundation for their different clinical efficacies.
