| 目的:探讨穿心莲内酯类药物对过氧化氢(H2O2)诱导的人脐静脉血管内皮细胞EA.hy926氧化损伤的影响。方法:采用1 mmol/L H2O2处理EA.hy926细胞4 h制备氧化损伤模型,将EA.hy926细胞分为对照组和模型组(H2O2组),加药组(穿心莲内酯、喜炎平、穿琥宁、炎琥宁)。CCK-8法检测加药后细胞的存活率;试剂盒检测细胞中超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量及细胞培养液中乳酸脱氢酶(LDH)的含量;2,7-二氯荧光素二乙酸酯探针检测细胞活性氧(ROS)生成。结果:与模型组比较,加药组中加入1 μmol/L穿心莲内酯与200 μg/ml喜炎平的细胞存活率显著增加(P < 0.05,P < 0.01);细胞内MDA含量显著降低(P < 0.05,P < 0.01);细胞培养上清中LDH释放量降低(P < 0.05,P < 0.01);SOD活性增加(P < 0.01);ROS水平下降(P < 0.01)。结论:穿心莲内酯和喜炎平可能是通过增强抗氧化酶SOD的活性,减少细胞内ROS的积累,进而减轻细胞内的脂质氧化,保护细胞膜结构的完整性,从而对H2O2诱导的血管内皮细胞氧化损伤产生良好的保护作用。 |

| Objective:To investigate the effects of andrographolide-based drugs on hydrogen peroxide (H?O?)-induced oxidative damage in human umbilical vein endothelial cells (EA.hy926).Methods: An oxidative damage model was established by treating EA.hy926 cells with 1 mmol/L H?O? for 4 hours. The cells were divided into three groups:control group, model group (H?O?-treated group), and drug-treated groups (treated with andrographolide, Xiyanping, Chuanhuning, and Yanhuning, respectively). Cell viability after drug administration was detected using the CCK-8 assay. Commercial kits were used to determine the activity of superoxide dismutase (SOD), the content of malondialdehyde (MDA) in cells, and the level of lactate dehydrogenase (LDH) in cell culture supernatants. Intracellular reactive oxygen species (ROS) generation was measured using the 2",7"-dichlorofluorescin diacetate probe.Results:Compared with the model group, the cells treated with 1 μmol/L andrographolide and 200 μg/ml Xiyanping in the drug-treated groups showed significantly increased viability (P < 0.05, P < 0.01), markedly decreased intracellular MDA content (P < 0.05, P < 0.01), reduced LDH release in cell culture supernatants (P < 0.05, P < 0.01), enhanced SOD activity (P < 0.01), and lowered ROS levels (P < 0.01).Conclusion: Andrographolide and Xiyanping may exert a favorable protective effect against H?O?-induced oxidative damage in vascular endothelial cells by enhancing the activity of the antioxidant enzyme SOD,reducing intracellular ROS accumulation, thereby alleviating intracellular lipid oxidation and preserving the structural integrity of cell membranes. |
