      目的:对葛根渣中资源性化学成分进行分析,并考察膳食纤维提取最佳工艺,以期为发现葛根渣资源价值和提升资源化利用效率提供参考。方法:通过苯酚—硫酸法、酸碱消煮法分别对葛根渣中的多糖和粗纤维含量进行测定分析,基于单因素试验采用α-淀粉酶、糖化酶和木瓜蛋白酶进行膳食纤维的制备,并测定膳食纤维的膨胀力,确定最佳工艺参数。结果:葛根渣中多糖含量为10.30%,粗纤维含量为15.58%;酶法制备膳食纤维的最佳工艺条件为:复合酶(α-淀粉酶、糖化酶)添加比例1:3、添加总量4%、酶解时间75 min、酶解温度65℃,蛋白酶添加总量为0.3%。结论:葛根渣中多糖、粗纤维等资源性化学成分丰富,所建立的膳食纤维提取工艺可行,研究结果可为葛根渣的转化增效及粗细高值化利用模式提供科学依据。

      Objective: By analyzing the resource based chemical components in pueraria root residue and examining the optimal process for extracting dietary fibe, we aim to provide reference for discovering the value of pueraria root residue resources and improving resource utilization efficiency. Methods: The content of polysaccharides and crude fibers in pueraria root residue was determined and analyzed by using the phenol sulfuric acid method and the acid-base digestion method, respectively. In addition, based on a single factor experiment with α-Amylase, saccharifying enzyme, and papain were used to prepare dietary fiber, and the swelling force of dietary fiber was measured to determine the optimal process parameters. Results: The polysaccharide content in pueraria root residue is 10.30%, and the crude fiber content is 15.58%; The optimal process conditions for enzymatic preparation of dietary fiber are as follows: the proportion of composite enzymes (α-Amylase and saccharifying enzyme) added is 1:3, the total amount added is 4%, the enzymatic hydrolysis time is 75 minutes, the enzymatic hydrolysis temperature is 65 ℃, and the total amount of protease added is 0.3%. Conclusion: The resource based chemical components such as polysaccharides and crude fibers in pueraria root residue are abundant, and the established dietary fiber extraction process is feasible. This research results can provide scientific basis for the transformation efficiency enhancement and high-value utilization mode of pueraria root residue.
