      目的:建立同时测定吴茱萸中去氢吴茱萸碱、柠檬苦素、吴茱萸碱、吴茱萸次碱的HPLC方法,测定不同产地吴茱萸药材有效成分含量及配比关系。方法:采用色谱柱为Dikmatech Diamonsil Plus C18(250×4.6 mm ,5 μm);流动相为乙腈(A)-0.02%磷酸溶液(B),梯度洗脱,紫外检测波长为215 nm;流速 1 mL?min-1,进样量为10 μL。结果:去氢吴茱萸碱、柠檬苦素、吴茱萸碱、吴茱萸次碱检测质量浓度范围分别为7.663~245.2、8.09~258.88、2.035~65.12、3.719~119μg?mL-1 (r≥0.9999),精密度、重复性、稳定性和耐用性试验中的RSD均小于3%(n=6),平均回收率在92.30%~106.9%,RSD均小于3%(n=6),不同产地去氢吴茱萸碱、柠檬苦素、吴茱萸碱、吴茱萸次碱配比关系存在差异。结论:该方法快速简便,分离效果好,重复性好,准确度高,可用于吴茱萸药材有效成分的含量测定和配比关系研究,含量配比关系的研究为其质量标准的制订提供了参考。
英文摘要:
      To develop an HPLC method for simultaneous determination of four active ingredients in Euodia rutaecarpa, including dehydroevodiamine, limonin, evodiamine, rutaecarpine and determine the contents and proportioning of the active ingredients in Euodia rutaecarpa. METHODS: The determination was performed on Dikmatech Diamonsil Plus C 18 column,the mobil phase was acetonitrile-buffer(0.02%phosphoric acid),gradient elution. UV detection wavelength was set at 215 nm and the flow rate was 1 mL?min-1. Sample size was 10 μL. RESULTS:The linear range of dehydroevodiamine, limonin, evodiamine and rutaecarpine were 7.663-245.2, 8.09-258.88 , 2.035-65.12and 3.719-119μg?mL-1 (r≥0.9999) respectively. RSDs of precision,repeatability,stability and durability tests were all less than 3%(n=6). The average recovery rate was 92.30%~106.9% , RSD is less than 3% (n=6),respectively. CONCLUSIONS: The method is proved to be accurate and valid,and can be used to study the contents and proportion relationship of active ingredients in Euodia rutaecarpa.The study of proportion relationship of active ingredients provide scientific basis for the formulation of quality standards.
