      目的 探究三草尿毒灵调控Shh/Gli信号通路抑制肾脏纤维化的作用与机制。方法:选取24只雄性BALB/c小鼠并随机分为假手术组、模型组、三草尿毒灵组以及CPN组,每组6只。制备单侧肾缺血再灌注损伤模型后第3天起,三草尿毒灵组每天给予三草尿毒灵汤剂(9g/kg)灌胃,CPN组每天给予CPN(5mg/kg)腹腔注射。术后第10天切除小鼠右侧肾脏,第11天处死小鼠,获取小鼠血清和肾脏进行肾功能检测、免疫组化、Western Blot和qRT-PCR分析。结果:与假手术组相比,模型组的血肌酐和尿素氮水平明显升高,而且相关纤维化指标的表达显著上调(P<0.05)。而三草尿毒灵与CPN均可明显保护小鼠肾功能,减轻肾脏纤维化(P<0.05)。进一步研究发现三草尿毒灵可以抑制肾脏Shh/Gli信号表达及成纤维细胞的活化。结论:三草尿毒灵可通过调控Shh/Gli信号通路的表达以减轻肾脏纤维化。
英文摘要:
      Objective: To explore the effect of Sancaoniaoduling in alleviating renal fibrosis by Shh/Gli signaling pathway. Methods: 24 Male BALB/c mice were randomly divided into Sham Group, unilateral renal ischemia/reperfusion injury (UIRI) Group, Sancaoniaoduling Group and CPN Group, each group with 6 mice. At 3 days after UIRI, mice were subjected to either daily intragastric administration of Sancaoniaoduling at 9 g/kg body wt. for 7 days or daily intraperitoneal injections of CPN at 5 mg/kg body wt. for 7 days. At 10 days after UIRI, mice were subjected to right kidney nephrectomy. At 11 days after UIRI, serum and kidney tissues were collected for further investigation, such as renal function assay, immunohistochemistry study, western blot and qRT-PCR analysis. Results: Compared with sham control, serum creatine and BUN level were significantly increased in the UIRI model and induced serious renal fibrosis (P<0.05). Both the Sancaoniaoduling and CPN treatment could protect renal function and reduce renal fibrosis in UIRI (P<0.05). In addition, Sancaoniaoduling could inhibit Shh/Gli signaling in renal tissues, which promote activation of renal interstitial fibroblast proliferation. Conclusion: Sancaoniaoduling may inhibits renal fibrosis by regulating Shh/Gli signaling pathway.
