      通过网络药理学研究射干-麻黄治疗COVID-19的成分、靶点及通路,研究其中的机制。在TCMSP数据库中检索射干、麻黄的化学成分以及靶点信息,在GeneCards数据库和OMIM数据库和NCBI基因数据库中筛选出疾病的靶点。在Bioinformatics Evolutionary Genomics 数据库中筛选出射干-麻黄作用于疾病的靶点,用String数据库进行PPI网络的构建,将PPI网络导入Cystoscap 3.7.2中,通过NetworkAnalyzer工具进行拓扑分析, 应用 Cytoscape 3. 7. 2 软件构建中药-成分-疾病-靶点网络图,应用R语言进行GO富集及KEGG富集分析。结果:射干-麻黄筛选获得40个活性成分,218个药物靶点,与COVID-19共同靶点48个;射干-麻黄中的鼠李黄素、异鼠李素成分在治疗COVID-19中起主要作用。射干-麻黄治疗COVID-19的主要靶点蛋白是IL6、CASP3、MAPK3、MAPK8、IL10、CCL2等。射干-麻黄中的有效成分主要通过对脂多糖反应、对细菌源分子反应、细胞对内毒素的反应、细胞对细菌源分子反应等生物过程作用于COVID-19,射干-麻黄治疗COVID-19的主要信号通路有AGE-RAGE信号通路、IL-17信号通路、恰加斯病(美国锥虫病)、利什曼病等。体现了射干-麻黄治疗COVID-19多成分、多靶点、多途径的特点。

      The components, targets and pathways of Shegan-ephedra in the treatment of COVID-19 were studied by network pharmacology, and the mechanism was studied. The chemical composition and target information of Radix Scutellariae and Ephedra were searched in TCMSP database, and the disease targets were screened out in GeneCards database, OMIM database and NCBI gene database. The target of Radix Rehmanniae-ephedra was screened out from Bioinformatics-Evolutionary Genomics database. The PPI network was constructed by String database, and the PPI network was introduced into Cystoscap 3.7.2. The topology analysis was carried out by NetworkAnalyzer tool, and the traditional Chinese medicine-component-disease-target network diagram was constructed by Cytoscape 3.7.2 software. GO enrichment and KEGG enrichment analysis were carried out by R language. Results: 40 active components, 218 drug targets and 48 common targets with COVID-19 were obtained by screening Shegan-Ephedra, and the rhamnetin and isorhamnetin components in Shegan-Ephedra played an important role in the treatment of COVID-19. The main target proteins of Shegan-ephedra in the treatment of COVID-19 are IL6, CASP3, MAPK3, MAPK8, IL10, CCL2 and so on. The active components in COVID-19,-Ephedra mainly act on the biological processes such as lipopolysaccharide reaction, bacterial molecular reaction, cell response to endotoxin, cell response to bacterial molecular reaction, and so on. The main signal pathways of COVID-19,-Ephedra treatment of COVID-19 are AGE-RAGE signal pathway, IL-17 signal pathway, Chagas disease (American trypanosomiasis), leishmaniasis and so on. It reflects the characteristics of multi-component, multi-target and multi-pathway in the treatment of COVID-19 with Shegan-Ephedra.
