目的 研究妊娠期糖尿病血糖控制不达标状态下和体外高糖环境下对Klotho基因表达的影响。 方法 选择妊娠期糖尿病患者103例,其中血糖控制不达标56例,血糖控制达标47例,采用ELISA检测血清Klotho蛋白水平并分析各组妊娠结局,同时在体外高糖环境下培养HEK293细胞,采用RT-PCR方法检测细胞Klotho基因表达水平。结果 血清α-Klotho 和β-klotho蛋白在妊娠期糖尿病血糖控制不达标组表达水平明显低于血糖控制达标组(p<0.05),此外,血糖不达标组剖宫产率、巨大儿发生率高于达标组(p<0.05)。RT-PCR结果表明高糖可诱导HEK293细胞α-Klotho 和β-klotho mRNA表达水平下降。结论 体内外高糖是导致α-Klotho 和β-klotho表达下降重要因素之一,妊娠期糖尿病治疗目标应以血糖达标最为获益。 |

Objective To investigate glucose effect on Klotho expression in vitro and in vivo. Methods 103cases of gestational diabetes mellitus were collected from JIANGXI MATERNAL AND CHILD HEALTH HOSPITAL, which were divided into two groups based on the glucose status. Serum Klotho was measured with ELISA kit. At the same time, HEK293 cell were cultured with high glucose exposure and RT-PCR was used to detect the Klotho mRNA expression under high glucose condition. Results Both α-Klotho and β-klotho were lower in gestational diabetes mellitus group with glucose control failure than gestational diabetes mellitus group with glucose control (p<0.05). In addition, the incidence of cesarean section and macrosomia in gestational diabetes mellitus group with glucose control failure was significantly high than gestational diabetes mellitus group with glucose control (p<0.05). RT-PCT results showed that high glucose downregulated the expression of Klotho mRNA in HEK293 cell line. Conclusions increase glucose directly affect the expression of Klotho in vitro and in vivo, the goal of gestational diabetes mellitus treatment is to maintain blood glucose levels achieving targets. |
