目的:研究蜂胶中短叶松素的制备工艺及含量测定方法,并测定和比较12个产地蜂胶中短叶松素的含量。方法: 采用溶剂提取、大孔树脂吸附及葡聚糖凝胶色谱和重结晶等方法进行分离纯化,根据理化性质和核磁数据对化合物进行结构鉴定为短叶松素;用HPLC法测定蜂胶中短叶松素含量,YMC C18柱(4.6mm×250mm,5μm),流速为1.0 mL.min-1,甲醇-0.1%磷酸溶液(60:40),检测波长:292nm。结果:所得短叶松素的纯度在98.5%以上,短叶松素在8.512~,425.6μg线性关系良好,平均回收率( n = 6) 分别为98. 60% ,RSD 分别为0.97%,12个产地的蜂胶中短叶松素以新疆、云南、陕西三地含量较高。结论:提取分离的短叶松素含量高,可作为含量测定用对照品,含量测定方法稳定、可靠,可作为蜂胶中短叶松素含量测定方法。 |

Objective: Method of preparing technique and determination of pinobanksin propolis on the content of the short and Ye Songsu were determined and compared in 12 areas in propolis.Method: By solvent extraction,macroporous resin adsorption and Sephadex gel column chromatography and recrystallization were used for separation and purification, according to the physicochemical properties and NMR data of compound structures were identified as pinobanksin; propolis pinobanksin content was determined by HPLC, YMC C18 column (4.6 mm x 250 mm, 5 m) at a flow rate of 1.0 mL.min-1 and methanol-0.1% phosphoric acid solution (60:40), the detection wavelength: 292nm. Result: The pinobanksin purity was above 98.5%, pinobanksin in 8.512 ~ 425.6 g good linear relationship. The average recovery rate (n = 6) were 98.60%, RSD were 0.97%, Pinobanksin 12 places in Xinjiang、Yunnan、Shanxi high propolis three local content.Conclusion: Extraction and separation of high content pinobanksin, as content was determined by standard, content determination method is stable, reliable, and can be as a determination method for propolis pinobanksin content |
