      目的:探讨补肾健脾中药复方对成骨细胞中ASK1、ERO1、Caspase-12调节作用机制。方法:将新西兰大白兔9只随机分为中药组、西药组、空白组三组,每组3只。三组分别用中药提取液、雌二醇混悬液按中药剂量和同体积(约10ml)蒸馏水灌胃,1月后抽取含药血浆。选用1日龄SD大鼠获取原代的成骨细胞体外培养,分为中药组、西药组、凋亡空白组、正常空白组。中药组、西药组分别给予对应的含药血浆培养;凋亡空白组和正常空白组均给予常规血清培养。24小时后采用检测ASK1、 ERO1和Caspase-12的含量。结果:在ASK1方面,中药组、西药组与凋亡空白组相比,差异有统计学意义(P<0.05);而且西药组比中药组更能显著降低ASK1 mRNA的表达,其差异有统计学意义(P<0.05)。在ERO1方面,中药组、西药组与凋亡空白组相比,差异有统计学意义(P<0.05),而且西药组比中药组更能显著降低ERO1蛋白的表达,其差异有统计学意义(P<0.05)。在Caspase-12方面,中药组、西药组与凋亡空白组相比,差异无统计学意义(P>0.05)。结论:补肾健脾中药复方通过ASK1、ERO1所介导的方式对成骨细胞的凋亡进行调控。
英文摘要:
      Objective:To explore effectsSof Chinese medicine for Bushenjianpi onSASK1, ERO1 and Caspase-12 in osteoblasts. Methods:9 New Zealand rabbits were randomly into the Chinese medicine group(n=3) ,the western medicine group(n=3) and control group(n=3) . The Chinese medicine group was given the intragastric administration with Chinese herbal compound.The western medicine group was given the alendronate sodium,control group was given distilled water.After treament for 1 month,the serum was separated from blood.1day old SD rats were used to culture osteoblasts ,which were divided into the Chinese medicine group,the western medicine group , apoptosis group and control group.The Chinese medicine group,the western medicine group were given the corresponding serum,other group were given distilled water.After 24h, GADD34、ERO1、IRE-1were examined. Results:Compared with the apoptosis group, the Chinese medicine group and the western medicine group could decrease the expression level of ASK1 and ERO1 in serum obviously(P<0.05). Compared with the Chinese medicine group in ASK1 and ERO1, the western medicine group could decrease the expression level obviously(P<0.05). But compared with the apoptosis group, there were no statistical significance in Caspase-12. Conclusion: Chinese medicine for Bushenjianpi is regulated by ASK1 and ERO1 in the way of apoptosis..
