| 摘要 目的 探讨口腔鳞状细胞癌(oral squamous cell carcinoma，OSCC)中PLK1（polo-like kinase 1，PLK1）蛋白表达与中心体扩增之间的相关性。方法 采用免疫组化SABC法检测10例正常口腔粘膜和47例OSCC中PLK1蛋白和γ-微管蛋白的表达情况，并用SPSS13.0统计软件分析两者之间的相关性。结果 PLK1蛋白和γ-微管蛋白在正常口腔粘膜组和OSCC组中的阳性表达率分别为0/%（0/10）、70.2%（33/47）和0/%（0/10）、76.6％（36/47），PLK1蛋白和γ-微管蛋白表达在正常口腔粘膜组和OSCC组之间比较差异均具有统计学意义（P＜0.05）。Spearman相关分析显示OSCC组织中PLK1蛋白与γ-微管蛋白表达之间具有正相关关系（r=0.305，P＜0.05）。结论 PLK1蛋白过表达可能是中心体扩增的潜在机制之一，PLK1蛋白过表达导致中心体扩增及细胞恶性增殖在OSCC发生中可能起一定作用。 |

| 【】Objective To study the correlation between polo-like kinase 1 protein and centrosome amplification in oral squamous cell carcinoma. Methods Formalin-fixed, paraffin-embedded tissues of 10 cases of oral normal mucosa and 47 cases of oral squamous cell carcinomas (OSCC). The expressions of PLK1 andγ-tubulin proteins in these tissues were investigated by using Immunohistochemistry. The correlation between PLK1 protein expression and centrosome amplification in OSCC was statistically analyzed by SPSS 13.0. Results The percentages of cases with up-regulated PLK1andγ-tubulin protein were 0%(0/10) or 70.2%(33/47)and 0%(0/10) or 76.6% (36/47) in normal oral mucosa and OSCC separately, the percentags of PLK1 and γ-tubulin protein over-expression in OSCC are significant higher than that in normal oral epithelial (P<0.05), a positive correlation could be found between the expressions of PLK1 and γ-tubulin protein in OSCC(r=0.305, P<0.05). Conclusion Over-expression of PLK1 protein is likely a contributing factor towards centrosome amplification in OSCC, which may play some roles in establish of OSCC. |
