      目的:观察右归饮对去卵巢血管钙化中谷氨酸受体变化的影响。方法:40只远交群大鼠(sprague Dawley,SD)雌性大鼠随机分为假手术组,模型组,低剂量组,高剂量组以及辛伐他汀组,采用华法令加维生素D3制备血管钙化模型,于造模后,低剂量组、高剂量组分别给予右归饮浓缩液5.35g·kg-1·d-1、16.05g·kg-1·d-1灌胃,辛伐他汀组予辛伐他汀混悬水溶液按照5.25mg·kg-1·d-1灌胃,假手术组和模型组给予等量生理盐水,共12W;于实验结束时采集血浆标本测定雌二醇以及血脂四项检测,苏木精—伊红染色法(HE染色)观察动脉钙化的情况以及应用免疫印迹试验检测N-甲基-D-天门冬氨酸受体(NMDAR,NR)1蛋白的表达。结果:模型组大鼠雌二醇、甘油三酯下降以及NR1蛋白表达降低(P﹤0.05),低密度脂蛋白升高(P﹤0.05);高剂量组以及低剂量组可提高雌二醇、甘油三酯含量并增加NR1蛋白表达(P﹤0.05),高剂量组和低剂量组以及辛伐他汀组可降低低密度脂蛋白的含量(P﹤0.05),辛伐他汀组对雌二醇无影响(P>0.05)。结论:右归饮可抑制去卵巢血管钙化的形成,可能与提高雌激素水平以及提高NR1蛋白的表达有关。
英文摘要:
      Objective To study the possible mechanism of Youguiyin in regulating glutamate receptors of vascular calcification in ovariectomized rats. Methods Totally forty SD rats were randomized into the sham group, model group, low-dose and high-dose group and simvastatin group, with in 8 each. The Artery calcification models were established by subcutaneously injecting warfarin and vitamin D3 for one week; the low-dose and high-dose group and simvastatin group was gavaged with 5.35g.kg-1, 16.05g.kg-1 Concentrated solution of Youguiyin,5.25mg.kg-1 suspensions of simvastatin respectively for 12 week,the ham group and model group were given equal amount of physiological saline;The contents of serum estradiol and lipid(including TC、TG、LDL、HDL) were detected in all groups,the HE staining of arterial calcification was observed through microscope and the expression of NR1 in arterial calcification were detected with western blotting.Results Compared with the sham group, the contents of estradiol and triglycerides and the expression of NR1 protein were significantly decreased in the model group(P <0.05), the content of low density lipoprotein was significantly increased (P <0.05),and the expression of NR1 in high-dose group was no significant difference between groups; compared with the model group, the content of estradiol,triglyceride levels protein expression of NR1 were significantly increased in the high dose group and low dose group(P <0.05)the content of LDL was significantly decreased in the high dose group and low dose group and simvastatin group;the content of estradiol in the simvastatin group,the content of Cholesterol and high-density lipoprotein were no differences among groups (P> 0.05).Conclusion Youguiyin may protect the arteries by inhibiting the formation of artery calcification,its mechanism may be relevant to enhancing estrogen levels and increasing the expression of NR1 protein.
