Objective: To observe the effect of different dose of ulinastatin (UTI) in hepatic and renal function on rats with paraquat (PQ) induced acute lung injury. Methods: 50 SD rats were randomly divided into 5 groups, normal control group (A), PQ poisoning group (B), low dose UTI treatment group (C), middle dose UTI treatment group (D), high dose UTI treatment group (E), 10 rats in each group. B, C, D, E group of rats were made into the poisoning model by intragastric PQ (80 mg/kg) administration, and same quantity of physiological saline was administered intragastrically in group A. 30 minutes after PQ administration, group C were intraperitoneally injected with UTI (100 kU/kg), group D were intraperitoneally injected with UTI (300 kU/kg), group E were intraperitoneally injected with UTI (600 kU/kg), and then the A,B group were injected same quantity of physiological saline. The levels of serum ALT, AST, BUN, Cr concentration of rats after 24 hours were measured, the pathological changes of lung tissues, lung injury score under the light microscope were observed. Results: Compared with B group, there were significant differences in C, D, E group with the serum ALT, AST, BUN, Cr concentration and lung tissue pathological score (P<0.05); and serum ALT, AST, BUN, Cr concentration had no significant difference between C, D, E group (P>0.05), there were significant differences in pathological lung tissue injury score (P>0.05). Conclusion: High dose UTI has strong protective effect on acute lung injury induced by paraquat-intoxicated in rats without significant effect on liver and renal function. |
