      目的:基于网络药理学方法探究黑地黄丸治疗肾性贫血的作用机制。方法:运用网络数据库对黑地黄丸成分及肾性贫血靶点进行检索筛选;运用Cytoscape软件,构建中药成分-靶点网络,运用STRING数据库构建黑地黄丸治疗肾性贫血的蛋白互作网络图,进行拓扑分析,找出核心靶点,最后用DAVID6.8数据库对药物-疾病交集靶点进行GO生物过程及KEGG通路富集分析。结果:在黑地黄丸中共筛选出45个与肾性贫血相关的有效成分和725个潜在作用靶点;拓扑分析得到12个核心靶点;GO生物功能分析得到129条富集结果,其中生物过程107条,细胞组成8条,分子功能14条,主要包括蛋白质结合、细胞外空间、细胞质、RNA聚合酶Ⅱ启动子转录的正调控等;KEGG通路分析得到3条通路,主要涉及HIF-1信号通路、PI3K-Akt信号通路、癌症通路等。结论:黑地黄丸治疗肾性贫血具有多成分、多靶点、多途径的特点,预测其主要活性成分、关键靶点和作用通路及生物过程为研究其作用机制和物质基础提供了理论依据。
英文摘要:
      Objective: To explore the mechanism of Heidihuang pills in treating renal anemia based on network pharmacology. Methods: The components of Heidihuang pills and targets of renal anemia were searched and screened by network database. The network of traditional chinese medicine components-target was constructed by using the Cytoscape software, and the protein interaction network diagram of Heidihuang pills in the treatment of renal anemia was constructed by using STRING database. The core target was found out by topological analysis. Finally, GO biological process and KEGG pathway enrichment analysis of drug-disease intersection target were carried out with DAVID 6.8 database. Results: 45 Active components and 725 potential targets related to renal anemia were screened out in Heidihuang pills; 12 core targets were obtained by topological analysis; 129 enrichment results were obtained by GO biological function analysis, including 107 biological processes, 8 cell compositions and 14 molecular functions, mainly including protein binding, extracellular space, cytoplasm, and RNA polymerase Ⅱ promoter transcription, etc. KEGG pathway analysis identified three pathways, mainly involving HIF-1 signaling pathway, PI3K-Akt signaling pathway, cancer pathway and so on. Conclusion: Heidihuang pills have the characteristics of multi-components, multi-targets and multi-pathways in the treatment of renal anemia. The prediction of its main active components, key targets, action pathways and biological processes provides a theoretical basis for studing its mechanism and material basis.
