      目的:基于网络药理学方法探究黑地黄丸治疗肾性贫血的作用机制。方法:运用网络数据库对黑地黄丸成分及肾性贫血(Renal anemia)靶点进行检索筛选。利用Cytoscape软件,构建中药成分-靶点网络。利用STRING数据库构建黑地黄丸治疗RA的蛋白互作网络图(PPI),拓扑分析出核心靶点。最后用DAVID 6.8数据库对药物-疾病交集靶点进行GO生物过程及KEGG通路富集分析。结果:在黑地黄丸中共筛选出45个与RA相关的有效成分和725个潜在作用靶点;拓扑分析得到12个核心靶点;GO生物功能分析得到129条富集结果,其中生物过程107条,细胞组成8条,分子功能14条,主要包括蛋白质结合、细胞外空间、细胞质、RNA聚合酶II启动子转录的正调控等;KEGG通路分析得到条通路,主要涉及HIF-1信号通路、PI3K-Akt信号通路癌症的通路等。结论:黑地黄丸治疗RA具有多成分、多靶点、多途径的特点,并预测其主要活性成分、关键靶点和作用通路及生物过程,为研究其作用机制和物质基础提供了理论依据。

      Objective: To explore the mechanism of Heidihuang Pills in treating Renal anemia based on network pharmacology. Methods: The components of Heidihuang Pills and targets of Renal anemia were searched and screened by network database. The network of components and targets of traditional Chinese medicine was constructed by using the software Cytoscape. The protein-protein interaction network map (PPI) of Heidihuang Pills for RA treatment was constructed using STRING database, and the core targets were topologically analyzed. Finally, DAVID 6.8 database was used to analyze GO biological process and KEGG pathway enrichment of drug-disease intersection targets. Results: A total of 45 RA-related active ingredients and 725 potential targets were screened out in Heidihuang Pills. Topological analysis revealed 12 core targets. GO biological function analysis showed 129 enrichment results, including 107 biological processes, 8 cell compositions and 14 molecular functions, mainly including protein binding, extracellular space, cytoplasm, positive regulation of RNA polymerase II promoter transcription, etc. The KEGG pathway analysis has resulted in three pathways, mainly involving HIF-1 signaling pathway, PI3K-Akt signaling pathway, cancer pathway, etc.Conclusion: Heidihuang Pill has the characteristics of multi-component, multi-target and multi-channel in the treatment of RA, and predicts its main active components, key targets, action pathways and biological processes, providing theoretical basis for studying its action mechanism and material basis.
